ОКСИДАЦІЙНІ ПОРУШЕННЯ ПРИ РОЗВИТКУ ЕКСПЕРИМЕНТАЛЬНОГО СТЕАТОГЕПАТИТУ

dc.descriptionIntroduction. Steatohepatitis is one of the leading pathologies which cause the damaging of hepatocytes and the development of an inflammatory process, which can subsequently lead to the development of cirrhosis. One of the main links in the pathogenesis of this process is the development of lipid peroxidation. Therefore, it is relevant to study the state of the pro- and antioxidant system in the process of developing of the experimental steatohepatitis. The Aim of the Study. Сhanges of the activity of superoxide dismutase, catalase and the content of lipid peroxidation products at developing of experimental steatohepatitis. Research Methods. The experiment was conducted on white mature male rats. The animals were divided into 4 groups: I – control, II – 7th day of steatohepatitis development, III – 14th day, IV – 28th day of the experiment. Accordingly, the content of diene and triene conjugates, catalase and superoxide desmutase activity were determined in the blood of animals of each group. The results were processed using BioStat Pro software. Normality was assessed with the Shapiro-Wilk test, and mean values were compared using Student's t-test. Data are presented as the arithmetic mean and standard fault (M ± SEM). Differences were considered statistically significant at p < 0.05. Results and discussion. The results of experiment showed an increasing of the level of lipid peroxidation products in the blood of animals during the development of experimental steatohepatitis, which was most pronounced on day 28. Such changes indicate increasing of the formation of active oxygen species development at experimental steatohepatitis, which cause to processes that can subsequently lead to damage to hepatocytes and the development of inflammation. The increase in the activity of superoxide desmutase and catalase indicates the formation of adaptive changes of antioxidant mechanisms in response to the development of oxidative stress during the development of experimental steatohepatitis. Conclusion: The development of experimental steatohepatitis is accompanied by an increasing of lipid peroxidation products in the blood of experimental animals and is accompanied by a change in the activity of the enzymatic link of antioxidant protection. This indicates that oxidative stress is an important link in the pathogenesis of experimental steatohepatitis.en-US

Стеатогепатит є одним з провідних патологій в уражені печінки, що супроводжується пошкодженням гепатоцитів та розвитком запального процесу, який в подальшому може привести до розвитку цирозу. Однією з провідних ланок патогенезу цього процесу є розвиток перекисного окиснення ліпідів. Тому актуальним є вивчення стану про- та антиоксидантної системи в процесі розвитку експериментального стеатогепатиту. Мета дослідження – визначити зміни активності супероксиддисмутази та каталази, вмісту продуктів перекисного окиснення ліпідів за умов розвитку експериментального стеатогепатиту. Методи дослідження. Дослідження було проведене на білих статевозрілих самцях щурів, яким моделювали експериментальний стеатогепатит. Тварини були розділені на 4 групи: І – контрольна, ІІ – 7-й день розвитку стеатогепатиту, ІІІ-14-й день, IV – 28-й день досліду. Відповідно в крові тварин кожної групи визначали вміст дієнових і трієнових кон’югатів, каталазну та супероксиддисмутазну активність. Отримані результати обробляли за допомогою програми BioStat Pro. Нормальність розподілу перевіряли за критерієм Шапіро-Вілка. Для порівняння середніх величин застосовували t-критерій Стьюдента. Результати подавали у вигляді середнього арифметичного та стандартної похибки (M±m). Статистично значущими вважали відмінності при p < 0,05. Результати й обговорення. Проведені дослідження показали зростання рівня продуктів перекисного окиснення ліпідів у крові тварин в процесі розвитку експериментального стеатогепатиту, що найбільш було виражене на 28 добу. Такі зміни вказують, що в печінці в гострий період розвитку експериментального стеатогепатиту наростає процес утворення активних форм кисню, які, у свою чергу, спричиняють каскад процесів, що в подальшому може приводити до пошкодження гепатоцитів та розвитку запалення. Зростання активності супероксиддисмутази та каталази свідчить про формування адаптаційних змін антиоксидантних механізмів у відповідь на розвиток оксидативного стресу при розвитку експериментального стеатогепатиту. Висновок: Розвиток експериментального стеатогепатиту супроводжується зростанням в крові експериментальних тварин рівня продуктів пероксидації ліпідів та супроводжується зміною активності ферментативної ланки антиоксидантного захисту. Це свідчить, що оксидативний стрес є важливою ланкою патогенезу експериментального стеатогепатиту.uk-UA
