СУРФАКТАНТНИЙ ПРОТЕЇН А1 (SP-A1) – МОЛЕКУЛЯРНИЙ БІОМАРКЕР УШКОДЖЕННЯ ЛЕГЕНЬ ПРИ ЕКСПЕРИМЕНТАЛЬНОМУ ЦУКРОВОМУ ДІАБЕТІ

dc.descriptionSUMMARY. Diabetes mellitus occupies one of the first places in the structure of endocrine diseases and affects many organs, including the lungs. Pulmonary surfactant, mostly surfactant protein A1, plays a leading role in the pathogenesis of respiratory diseases. There is insufficient data on the use of serum surfactant protein A1 as a potential biomarker of lung damage in the scientific literature. The aim – to evaluate the informativeness of surfactant protein A1 content in blood serum as a prognostic biomarker of lung injury in experimental diabetes mellitus. Material and Methods. A model of diabetes mellitus, which was reproduced by intraperitoneal injection of streptozotocin by "Sigma" company (USA), diluted in 0.1 M citrate buffer with a pH of 4.5, at a rate of 60 mg/kg body weight. The control group of animals received an intraperitoneal injection with an equivalent dose of 0.1 M citrate buffer solution with a pH of 4.5. Serum levels of SP-A1 were determined by rat enzyme-linked immunosorbent assay Rat ELISA Kits (Elabscience, USA) according to the manufacturer's instructions 14, 28, 42 and 70 days after streptozotocin injection. Results. Conducted biochemical analysis showed an increase in the levels of SP-A1 in blood serum in animals with diabetes mellitus at all stages of the experiment. In particular, serum SP-A1 levels increased by 7.9 % in 14 days, 49 % in 28 days, 69.5 % in 42 days and 91.6 % in 70 days compared to the control group of animals. Conclusions. During the entire study period, experimental diabetes mellitus is accompanied by an increase in surfactant protein A1 levels in blood serum and can be considered as a molecular biomarker of lung damage in this pathology.en-US
dc.descriptionРЕЗЮМЕ. Цукровий діабет посідає одне з перших місць у структурі ендокринних захворювань і вражає багато органів, у тому числі й легені. Важливою ланкою в патогенезі захворювань органів дихання є стан сурфактанту легень і зокрема сурфактантного протеїну А1 (SP-A1). У науковій літературі недостатньо даних щодо використання сироваткового SP-A1 як потенційного біомаркера ушкодження легень. Мета – оцінити інформативність вмісту сурфактантного протеїну А1 в сироватці крові в якості прогностичного біомаркера ушкодження легень при експериментальному цукровому діабеті. Матеріал і методи. Модель цукрового діабету відтворювали шляхом внутрішньоочеревинного введення білим щурам стрептозотоцину фірми «Sigma» (США), розведеного в 0,1 М цитратному буфері з рН 4,5, з розрахунку 60 мг/кг маси тіла. Контрольній групі тварин внутрішньоочеревинно вводили еквівалентну дозу 0,1 М цитратного буферного розчину з рН 4,5. У сироватці крові визначали вміст SP-A1 за методом імуноферментного аналізу з використанням наборів Rat ELISA Kits (США) через 14, 28, 42 і 70 діб після ін’єкції стрептозотоцину. Результати. Проведені біохімічні дослідження сироватки крові показали, що у тварин з цукровим діабетом спостерігається підвищення рівня SP-A1 на всіх етапах експерименту. Зокрема, вміст SP-A1 у сироватці крові зріс через 14 діб на 7,9 %, через 28 діб на 49,0 %, через 42 доби на 69,5 % і через 70 діб на 91,6 %, порівняно з показниками контрольної групи тварин. Висновки. Експериментальний цукровий діабет протягом усього періоду дослідження супроводжується підвищенням у сироватці крові вмісту сурфактантного протеїну А1 і може розглядатись в якості молекулярного біомаркера легеневого ушкодження при даній патології.uk-UA
