According to information received the study drug has a positive effect on the course of spontaneous pain response in rats and has a pronounced analgesic activity.Keywords: glucosamine, ketoprofen, a cream-gel, a spontaneous reaction of pain, analgesic activity.Introduction. Osteoarthritis (OA) - a chronic degenerative joint disease, characterized by progressive destruction of articular cartilage, proliferative reaction of cartilage and bone and accompanied by reactive synovitis. The disease is one of the most common pathology in the structure of musculoskeletal system: OA prevalence in the population is 14% of the adult population. In order to improve the quality of life of the patient rational use of drugs that could adjust the pain, such as narcotic analgesics and non-steroidal anti-inflammatory drugs (NSAIDs). But only eliminate pain in osteoarthritis is not enough, because the disease is in violation of cartilage metabolism. So today, on the recommendation EULAR symptomatic slow-acting drugs (glucosamine, chondroitin sulfate, diatsereyin and еtс.) are used on the list of recommendations for the treatment of patients with OA. A combination of the above two groups can influence both the pain of this pathology and the metabolism of articular cartilage. Pain in osteoarthritis is permanent, so pain relief is needed dosage form that would have convenient use. Certain topical dosage forms such as gel, cream so cream-gel have this properties. The aim of this research was to study the analgesic properties of new combined drug - a combination of glucosamine and ketoprofen, the ratio of 2.5:1 in the form of a cream-gel (G/K cream-gel) during playback spontaneous pain reaction in rats on a background of acute arthritis of knee.Methods. The model of acute arthritis of the knee joint on the right rear paw by intraarticular injection of 25 ml of 2% solution λ-karaheninu (Sigma, USA), produced in aseptic saline was used. Measuring the intensity of spontaneous pain reaction carried out at 1, 3 and 5 day of study, each animal was placed before fixing on the "tester disability" camera - Incapacitance Tester MkV. Through measurements index disability (ID) and analgetic activity (AA) were fixed. Drugs of comparison were Glucosamine cream gel, Chondroxid and Fastum gel.Results and discussion.The results of the impact studies G/K cream gel on the intensity of spontaneous pain reaction in rats with acute gonarthritis show that the studied combination has a strong analgesic effect, since significantly increases ID during the period of observation. This causes the following indicators AA, which was the highest in the present study: as of 1 day research - 41.79%, 3 night - 47.92% and 62.23% 5 night that expressive actions since 3 days significantly exceeded preparations comparison. High levels of AA study drug because the combination of glucosamine and ketoprofen has both analgesic and anti-inflammatory effects and ability to improve the metabolism of articular cartilage. Drugs of comparison Glucosamine cream gel, Chondroxid had much lower levels of analgetic activity. The analgesic activity of Fastum gel though significantly different from the previous two, but was lower than the study combination.Conclusions1. Results of the experiment showed that the combination of glucosamine with ketoprofen in the form of a cream-gel with cutaneous application reveals pronounced analgesic effect in conditions of spontaneous pain reaction in rats. 2. According to the degree of influence of analgesic combination drug significantly exceeds analgesic activity compared Fastum gel on 3 and 5 days of study, and other reference samples throughout the all research.uk-UA
